There are plenty of difficult enemies and optional mini-bosses for you to take down in RE4 Remake. And while facing them is a dangerous ordeal, it can be very rewarding since they almost always drop Gemstones as loot.

For example, mini-bosses such as Brutes or Chainsaw Villagers have a 100% drop rate for Gemstones. Moreover, other difficult enemies such as Armadura Knight, which is a parasite-infested armored knight, and Novistador, which are the bat-like monsters that can either fly or camouflage themselves, will always drop a Gemstone.
