To use it, first, you need to fill up your focus bar, which is in the upper right corner of your screen (The yellow bar). Once it’s full, press the L3 and R3 buttons of your dual sense controller together to generate an epic shockwave around you, dealing massive damage.

If you’re wondering if you could miss out on unlocking this ability or finding out about it too late, then don’t worry because the game prompts you to use it in one of the main missions.
Specifically, you need to use the Mega Venom Blast in the 16th mission in the main story. The mission is called ‘Funky‘.
How To Fill Up The Focus Bar
Knowing how to fill up the focus bar is key if you want to perform the Mega Venom Blast often. Although pummeling enemies is the obvious way to fill it up, you can fill it faster by performing combos when fighting. Longer combos will contribute more to the focus bar.
Also, an additional way to fill up the focus bar is to dodge or parry as many enemy attacks as possible. Therefore, you need to be good with both offense and defense if you want to fill up the focus bar fast.
When To Use The Mega Venom Blast
